DM-58 LEO II Lyse is a hemolysing reagent used by Mindray BC 5800 hematological analyzer for the determination of lymphocyte cell count, monocyte cell count, eosinophil cell count, neutrophil cell count.
SUMMARY AND PRINCIPLE
A sample volume of a whole blood specimen is aspirated into the analyzer where a portion of it is automatically diluted with DM-58 LEO I Lyse and DM-58 LEO II Lyse. The red blood cells are hemolysed and all white blood cells selectively loose part of their cytoplasm and shrink in size. From the 4Diff reaction chamber the cell suspension through the flowcell along with DM-58 D Diluent. The two fluids flow as independent, concentric streams (no mixing), with the DM-58 D Diluent stream encasing the sample stream. The sample is then analyzed by two-angle laser light scattering detection using a laser diode. The cells in the suspension are classified into red blood ghosts, lymphocytes cells (LYM), monocytes cells (MONO), eosinophil cells (EO) and neutrophil cells (NEU).
