INTENDED USE
DiaLP reagent kits are applied in public health and clinical microbiological laboratories for the qualitative demonstration of Legionella pneumophila chromosomal DNA in liquid microbiological samples. DiaLP kit is a test for the detection of pathogenic Legionella DNAs identical to those of the most frequently examined Legionella pneumophila strains. Bacterial DNA is identified by real-time PCR amplification method.
TEST PRINCIPLE
The principle of the test is a real-time PCR process directed by specific primer-probe pairs. These oligonucleotides are specific to unique sequencesencoding both for pathogeneity islands of Legionella (pathogenic factor namedmacrophage infectivity potentiator - MIP) and conserved 16s rRNA regions ofLegionella pneumophila species. The primer system attached to the 5? regions ofORF (Open Reading Frame) gives rise to the amplification of single copy gene sequences together with the upstream regulatory elements. This amplificationsystem results in the detection of species-specific and virulence determiningregions of bacterial germs in one PCR reaction.
