SUMMARY AND PRINCIPLE
A sample volume of a whole blood specimen is aspirated into the analyzer where a portion of it is automatically diluted with Diastromlyser-SYS-FDI and Diastromlyser-SYS-FDII. The red blood cells are hemolysed and all white blood cells selectively loose part of their cytoplasm and shrink in size. From the 4Diff reaction chamber the cell suspension flows through the flowcell along with Diaton-SYS Diluent. The two fluids flow as independent, concentric streams. The Diaton-SYS Diluent stream is encasing the sample stream. The sample is then analyzed by two-angle laser light scattering detection using a laser diode. The cells in the suspension are classified into red blood ghosts, lymphocyte cells (LYM), monocyte cells (MONO), eosinophil cells (EO) and neutrophil cells (NEU).
