Baso Dil. is used by the Technicon H*1, H*2, H*3 hematological analyzer red blood cell hemolysing reagent for the assay of basophils, mononuclear cells, and polymorphonuclear cells.
SUMMARY AND PRINCIPLE
When the EDTA anticoagulated whole blood sample is mixed with Baso Dil. reagent, the red blood cells are hemolyzed and the cytoplasm is stripped from all white cells except basophils. The sample is then analyzed by two-angle laser light scattering detection using a laser diode. The white cells are classified into three categories: basophils, mononuclear (MN) cells, and polymorphonuclear (PMN) cells. A constant volume of the cell suspension from the Baso reaction chamber passes through the flowcell along with sheath reagent. The two fluids flow as independent, concentric streams (no mixing), with the sheath reagent stream encasing the sample stream.
